Instrument training - specific to each instrument

Each instrument in our core facility requires specific training by one of our staff before a user can be allowed to operate it by themselves. The exact length of training varies due to the complexity of the instrument and the previous experience of the new user. Training is charged at the the instrument's routine hourly rate plus a per hour staff labor fee (training rates for all of our instruments can be found in iLab). The staff member makes the determination as to when the user is ready to operate an instrument by themselves.

Training is often 1-2 sessions of 1.5 hours each. We limit the number of trainees to a maximum of two individuals from the same lab. We teach the trainee how to get the most of the instrument and often make suggestions about sample prep, alternate techniques, controls, etc.

BioTek Cytation 1 plate reader

The Cytation 1 is a key component of the Seahorse XFe96 plate reader. This system is capable of fluorescence detection and high contrast brightfield imaging and includes Gen5 Image+ Software, 4x objective, DAPI filter cube and 365 nm LED light source for DAPI with the ability to provide phenotypic and quantitative data in a single experiment

Applications include; cell migration and invasion assays, live cell imaging, label-free cell counting, multi-mode reader applications (including; immunofluorescence, phenotypic assays, ELISA plate reader)

Agilent Seahorse XFe96 Analyzer

Agilent Seahorse XFe96 Analyzers measure the oxygen consumption rate (OCR) and extracellular acidification rate (ECAR) of live cells in a 96-well plate format. OCR and ECAR rates are key indicators of mitochondrial respiration and glycolysis as well as ATP production rate. Together these measurements provide a systems-level view of cellular metabolic function in cultured cells and ex vivo samples.

Standard assays include; real-time ATP rate, cell mito stress test, substrate oxidation stress test, glycolytic rate test, palmitate (fatty acid) oxidation stress test, mito fuel flex test, human T-cell activation, glycolytic stress test.

User training, staff-assisted use, or unassisted use are available for this instrument.

Amnis/Luminex ImageStreamX Mk II - imaging flow cytometer

The ImageStreamX Mk II is a flow cytometer which uses digital cameras to record fluorescence images of each cell from suspension, instead of recording only fluorescence intensity from each cell. Because these images record fluorescence location -- co-localization, translocation, and morphology can now be measured for large numbers of individual cells -- making even rare event analysis possible. Four lasers (405nm, 488nm, 561nm, 642nm) enable 10 fluorescence channels as well as classic brightfield channels. Sophisticated image analysis algorithms allow for discrimination of cellular processes with statistical significance. Can be used to determine colocalization, translocation, and morphology for large numbers of cells, with the possibility of rare event analysis.

Cytek Aurora - spectral flow cytometer (5 lasers)

Includes five lasers (355nm, 405nm, 488nm, 561nm, 640nm), three scattering channels, and 64 fluorescence channels. This instrument is able to run complex multi-color panels (40+ colors). It is used primarily for deep immunophenotyping assays. The instrument includes a 96 well plate loader to enable high-throughput flow cytometry. The instrument is available on FluoroFinder.
