Agilent Seahorse XFe96 Analyzer
Agilent Seahorse XFe96 Analyzers measure the oxygen consumption rate (OCR) and extracellular acidification rate (ECAR) of live cells in a 96-well plate format. OCR and ECAR rates are key indicators of mitochondrial respiration and glycolysis as well as ATP production rate. Together these measurements provide a systems-level view of cellular metabolic function in cultured cells and ex vivo samples.
Standard assays include; real-time ATP rate, cell mito stress test, substrate oxidation stress test, glycolytic rate test, palmitate (fatty acid) oxidation stress test, mito fuel flex test, human T-cell activation, glycolytic stress test.
User training, staff-assisted use, or unassisted use are available for this instrument.

Amnis/Luminex ImageStreamX Mk II - imaging flow cytometer
The ImageStreamX Mk II is a flow cytometer which uses digital cameras to record fluorescence images of each cell from suspension, instead of recording only fluorescence intensity from each cell. Because these images record fluorescence location -- co-localization, translocation, and morphology can now be measured for large numbers of individual cells -- making even rare event analysis possible. Four lasers (405nm, 488nm, 561nm, 642nm) enable 10 fluorescence channels as well as classic brightfield channels. Sophisticated image analysis algorithms allow for discrimination of cellular processes with statistical significance. Can be used to determine colocalization, translocation, and morphology for large numbers of cells, with the possibility of rare event analysis.
Please acknowledge NIH award S10 OD028466 in publications which use data from this instrument, which was secured by the diligent work of Dr. Deepta Bhattacharya.
To use this instrument please contact the core via email to set an appointment.

Cytek Aurora - spectral flow cytometer (5 lasers)
Includes five lasers (355nm, 405nm, 488nm, 561nm, 640nm), three scattering channels, and 64 fluorescence channels. This instrument is able to run complex multi-color panels (40+ colors). It is used primarily for deep immunophenotyping assays. The instrument includes a 96 well plate loader to enable high-throughput flow cytometry. The instrument is available on FluoroFinder.
User training, staff-assisted use, or unassisted use are available for this instrument. The staff contact for this instrument is Patricia Gundy - pgundy@email.arizona.edu

BD FACSCanto II - flow cytometer
The FACSCanto II is equipped with 3 lasers (405nm, 488nm, 633nm), 8 fluorescence channels and 2 channels for Forward Scatter and Side Scatter. This instrument is an analysis instrument only. The FACSCanto II is best for DNA quantification in mammalian, plant or yeast cells.

BD FACSAria III - cell sorter and flow cytometer
The FACSAria III is equipped with 5 lasers (355nm, 405nm, 488nm, 561nm, 633nm), 17 fluorescence channels and 2 channels for Forward Scatter and Side Scatter. It is a state of the art cell sorter capable of physically separating 4 populations simultaneously. Sorted cells can be collected into tubes, plates, or onto slides. Typically staff will perform startup, QC, and sorting, but highly experienced LSR users may qualify to receive training in how to operate the FACSAria III unassisted.

BD LSR II - flow cytometer
The LSR II is equipped with 5 air-cooled lasers (355nm, 405nm, 488nm, 532nm, 640nm), 16 fluorescent channels and 2 channels for Forward Scatter and Side Scatter. It is a state of the art digital flow cytometer, containing octagon and trigon optical arrays, allowing increased signal sensitivity. This is a train-to-use instrument, contact the core staff for more information about training.
